Abstract
Background: Hereditary thrombophilias, such as deficiencies of natural
anticoagulants, as well as common thrombotic genetic mutations are well
documented as causes of recurrent spontaneous abortion. _
Objective: This study aimed to screen for both common thrombotic genetic
mutations and coagulation inhibitors deficiency related 'to recurrent
pregnancy loss among Sudanese women. ‘
Materials and methods: the study is a case control study, included 100
Sudanese women with unexplained recurrent pregnancy loss, and 50 healthy,
age-matched reproductive Sudanese women as a control group. Two blood
samples were collected fiom each subject, one in 3.2% tri-sodium citrate for
coagulation investigations and the other in ethylene diamine tetra acetic acid
(EDTA) for molecular analysis. _
Protein C, protein S, and antithrombin HI were measured by chromogenic
assay. Genomic DNA was extracted from EDTA samples and analysed for
the presence of factor V Leiden mutation, prothrombin G20210A mutation,
and methylene tetrahydrofolate reductase mutation using polymerase chain
reaction- restriction fiagment polymorphism (PCR-RFLP).
