Interleukin-10 (1082G/A) Polymorphisms and Susceptibility to Adult Acute Lymphocytic Leukemia in Sudanese Population

Abstract

Abstract Background: Interleukin-10 (IL-10) is a multifunctional cytokine with both immunosuppressive and anti-angiogenic functions and may have both tumor-promoting and -inhibiting properties. We examined the association between a single nucleotide polymorphism (SNP) in IL-10 -1082G/A (rs1800896) in Sudanese adult acute lymphoid leukemia (ALL) patients and to assess the association between polymorphisms in IL-10 -1082G/A (rs1800896) and the immunophenotype (B-ALL and T-ALL), age groups and gender in Sudanese adult patients with ALL. Methods: A total of 50 adult patients with acute lymphoid leukemia and 50 control subjects were enrolled in this study. Blood samples were collected from all patients in EDTA containing tubes. Genomic DNA was extracted from all blood samples using salting out method. The genotypic variants of IL-10 (-1082G/A) polymorphism were detected by allele specific-PCR. Results: We found that (12%) of patients have homogenous GG genotype, (80%) have heterogeneous GA genotype and (8%) have AA genotype. GA genotype was significantly associated with higher risk of ALL, GG and AA protective from this disease for adult. there is no association between IL-10 (-1082G/A) polymorphism and adult ALL immunophenotypes, gende rand age group ,.Conclusion: Our study concluded that GA genotype of IL-10 -1082G/A polymorphism is a risk factor for adult ALL. But GG and AA genotyps are protective. No association between IL-10 (-1082G/A) polymorphism and ALL immunophenotype, gender and age group. الخلاصة الخلفية: Interleukin-10 (IL-10) عبارة عن سيتوكين متعدد الوظائف له وظائف مثبطة للمناعة ومضادة لتولد الأوعية وقد يكون له خصائص تعزيز الورم وتثبيطه. قمنا بفحص العلاقة بين تعدد أشكال النوكليوتيدات المفردة (SNP) في IL-10-1082G / A (rs1800896) في مرض سرطان الدم الليمفاوي الحاد في السودان (ALL) وتقييم العلاقة بين تعدد الأشكال في IL-10-1082G / A (rs1800896)والنمط المناعي (B-ALL و T-ALL) والفئات العمرية والجنس في المرضى السودانيين البالغين المصابين بـ ALL. الطريقة:تم تسجيل ما مجموعه 50 مريضًا بالغًا مصابًا بسرطان الدم الليمفاوي الحاد و 50 شخصًا من المجموعة الضابطة في هذه الدراسة. تم جمع عينات الدم من جميع المرضى في أنابيب تحتوي على EDTAتم استخراج الحمض النووي الجيني من جميع عينات الدم باستخدام طريقة التمليح. تم الكشف عن المتغيرات الوراثية لتعدد الأشكال IL-10 (-1082G / A) بواسطة PCR النوعي للأليل. النتائج:وجدنا أن (12٪) من المرضى لديهم تركيب وراثي GG متجانس،و (08٪) لديهم نمط وراثيGA غير متجانس و (8٪) لديهم نمط وراثي AAارتبط النمط الجيني GA بشكل كبير مع ارتفاع مخاطر الإصابة بـALL ،أما AAو GG يشكلون حماية من هذا المرض للبالغين ولا يوجد ارتباط بينIL-10 (-1082G / A) تعدد الأشكال والبالغين في جميع الأنماط المناعية والجنس والفئة العمرية. الاستنتاج: خلصت دراستنا إلى أن النمط الجيني GA لـ IL-10 -1082G / A تعدد الأشكال هو عامل خطر بالنسبة للبالغين ALL وال AA , GG هو عامل حماية. لا يوجد ارتباط بين تعدد الأشكال IL-10 (-1082G / A) وجميع الأنماط المناعية والجنس و الفئة العمرية.
