Loop-Mediated Isothermal Amplification Assay (LAMP) for the Diagnosis of Malaria infection Compared with the Microscopy, RDT and PCR in Khartoum ,state, Sudan

Abstract

Abstract Background: Recently described loop-mediated isothermal polymerase chain reaction (LAMP) for molecular detection of Plasmodium falciparum was compared with microscopy, PfHRP2-based rapid diagnostic test (RDT), and nested polymerase chain reaction (PCR). Microscopy as the “gold standard” in 50 samples . Low parasite density malaria infections are difficult to detect Methods: 50 blood sample were collected from symptomatic patient, each sample was diagnosed by LAMP, microscopy, PfHRP2- based rapid diagnostic test (RDT) and nested polymerase chain reaction (nPCR), using microscopy as the gold standard Results: Using microscopy as the gold standard technique , the sensitivity RDT was 47% and the specificity was 24%, respectively. LAMP assay was 95% sensitive and 96.7% specific compared to nPCR ( 65% sensitivity and 60% specificity). Conclusions: This study further validates LAMP as an alternative molecular diagnostic tool ,which can be used in the diagnosis of early infections of malaria cases and together with microscopy can also be used as supplementary methods for clinical and epidemiological use. الخلاصة خلفية: للكشف الجزيئي عن طفيل المتصورة المنجيلية مع المجهر (LAMP) تمت مقارنة تقنية ال والمجهر كمعيار (PCR), اختبارات التشخيصالسريع)، وال ) ICT ( PfHRP الضوئى ، و( 2 قياسي " في 50 عينة. الطريقة: تم جمع 50 عينة دم من مرضى ذوي أعراض. تم تشخيصكل عينة بواسطة المجهري وذلك ، (nPCR) اختبارات التشخيصالسريع) والبلمرة المتداخلة ) ICT الضوئى ،وبواسطة ال باستخدام المجهر كطريقة فحصمعيارية. 47٪ ، وكانت النوعية ٪ 24 على ICT النتائج: استخدام المجهر كتقنية قياسية ، وكانت لل حساسية 65 ٪ و 60 ٪ نوعية nPCR 95٪ حساسية و 97 ٪ مع مقارنة ال LAMP التوالي. كان لل . الخلاصة : كأداة التشخيصالجزيئي البديلة، والتي يمكن أن تستخدم في LAMP تؤكد هذا الدراسة ان الل تشخيصالأمراضفي وقت مبكر من حالات الإصابة بالملاريا وجنبا إلى جنب مع المجهرالضوئي ويمكن أيضا أن تستخدم كوسيلة تكميلية للاستخدام السريري والوبائي
