ABSTRACT The present study was aimed at assessing the physiological and histological changes in rats following experimental induction of diabetes mellitus-like disease through injection with streptozotocin (STZ). Twenty healthy male rats (Wistar strain, weight 353±90 g, 3 months old) were kept in under a 12/12 h dark-light cycle, at 23.4ºC. They had free access to normal rat chow and tap water. These animals were divided in two groups injected once with either STZ (60 mg/kg, treatment group, 12 animals) or saline (same volume as STZ, control group, 8 animals). Then animals were monitored for 8 months (long-term diabetes model). Behavioural studies (hole board and pole tests) and histological tests were performed, and physiological parameters monitored (glycaemia, body weight). Results of these two groups were analysed using Mann-Whitney U-test, and difference were significant for P<0.05. Behavioural tests’ results revealed that diabetic animals displayed, compared to control group: (i) decreased head-dipping episode number (p<0.05) and increased latency to the first grooming episode (p<0.001); (ii) decreased total locomotor activity and pole time (38 time pole/s, P <0.001), as well increased t-turn time (45%, P <0.01). Also compared to control animals, histological studies in diabetic animals revealed: (i) an increased meningeal thickness and a higher density of circulating cells in the meninges (p<0.05); (ii) mitochondrial alterations such as swelling and/or vacuolation in neuronal cells; (iii) neuronal cell apoptosis; (iv) microglia-like cells and myelin disturbances in the neuropil; as well as (v) numerous activated brain macrophages. Glycaemia was also higher, and body weight lower, in the diabetic group. In conclusion, diabetes mellitus-like disease was marked by cognitive and motor changes possibly due to damage and loss in neuronal populations emerging partly from physiology alterations characteristic of the disease. مستخلص البحث هدفت الدراسة الحالية إلى تقييم التغيرات الفسيولوجية والنسيجية للفئران بعد التحريض التجريبي لمرض السكري الشبيه بالسكري عن طريق الحقن بالستربتوزوتوسين (ستز). تم الاحتفاظ ب 20 جرذان ذكور صحية (سلالة ويستار، الوزن 353 ± 90 غرام، 3 أشهر من العمر) تحت دورة ضوء مظلمة 12/12 ساعة، عند 23.4 درجة مئوية. كان لديهم حرية الوصول إلى تشو الفئران العادية والمياه الصنبور. تم تقسيم هذه الحيوانات إلى مجموعتين تم حقنهما مرة واحدة إما باستخدام ستز (60 ملغم / كغم، مجموعة معالجة، 12 حيوان) أو محلول ملحي (نفس حجم ستز، مجموعة السيطرة، 8 حيوانات). ثم تم رصد الحيوانات لمدة 8 أشهر (نموذج السكري على المدى الطويل). أجريت الدراسات السلوكية (لوحة حفرة واختبار القطب) والاختبارات النسيجية، ورصدت المعلمات الفسيولوجية (سكر الدم، وزن الجسم). تم تحليل نتائج هاتين المجموعتين باستخدام مان-ويتني U-تيست، وكان الفرق معنويا ل P <0.05. أظهرت نتائج الاختبارات السلوكية أن الحيوانات المصابة بالسكري تظهر مقارنة مع المجموعة الضابطة: (1) انخفاض عدد حلقات غمس الرأس (p <0.05) وزيادة زمن الانتقال إلى حلقة الاستمالة الأولى (p <0.001).
