Comparison of Gum Arabic for Use as Mounting Media for Arthropod and Protozoa cyst in Stool Sample

Abstract

Abstract Background The mounting medium is the solution in which the specimen is embedded. It may be liquid, gum or resinous, soluble in water, alcohol or other solvents. The main purpose of mounting media is to physically protect the specimen from damage, air drying effect, and fading. This study aim to assess the use of Gum Arabic as Mounting media for Arthropod and Protozoa cyst in stool sample. Method In this experimental study a fifty entomological items were mounted by 2 different mounting techniques(Puri’s media and DPX). & we use Hoyer’s media and Glycerin jelly for mounting protozoa cyst. Result We found that Puris technique provided excellent morphological details and clear items also there were no air bubbles. DPX technique showed air bubbles especially in adult insects. It was found in glycerol jelly mounted slides the morphology of the parasite were excellent and there were no air bubble, compared with hayer’s where parasite morphology appeared faint. We concluded that Puri’s media is best used for insect mounting. And Glycerol jelly monted specimen give the best result.

Conclusion We concluded that Puri’s media is best used for insect mounting. And Glycerol jelly mounted specimen give the best result.

الخلاصة

وسيط التركيب هو المحلول الذي يتم تضمين العينة فيه. قد يكون سائلًا أو صمغًا ,قابل للذوبان في الماء أو كحول أو مذيبات أخرى. الغرض الرئيسي من تركيب الوسائط هو الحماية المادية للعينة من التلف وتأثير تجفيف الهواء والتلاشي. تهدف هذه الدراسة إلى تقييم استخدام الصمغ العربي كوسيط لحفظ مفصليات الأرجل وكيس البروتوزوا في عينة البراز.

في هذه الدراسة التجريبية ، تم تحضير خمسين عنصرًا من الحشرات باستخدام تقنيتي تركيب مختلفتين (وسائط Puri وDPX) ونستخدم وسائط Hoyer وهلام الجلسرين لتركيب كيس الطفيليات.

وجدنا أن تقنية Puris قدمت تفاصيل مورفولوجية ممتازة وعناصر واضحة كما لم تكن هناك فقاعات هواء. أظهرت تقنية DPX فقاعات هواء خاصة في الحشرات البالغة. وُجد في شرائح الجلسرين المركبة على شكل هلام ، أن شكل الطفيلي كان ممتازًا ولم تكن هناك فقاعة هواء ، مقارنةً بالشرائح التي ظهرت فيها الطفيليات باهتة وجد ان وسيط Puris هو الافضل لحفظ عينة الحشرات، كما يعد وسيط الجليسرين الافضل للطفيليات. هذه الدراسة خلصت الى ان استعمال وسيط Puris هو الافضل لحفظ الحشرات كما ان هلام الجلسرين هو الافضل لحفظ الطفيليات.
