T Background: Cryptococcus neoformans is an important fungal pathogen that is recognized for its ability to cause subacute CNS disease and Yeast are ubiquitous in the environment and often are part of the microbiota of both humans and animals. Cryptococcosis has been considered a sporadic infection and varies widely with geographic location according to the etiologic agent. C. neoformans is the most common cause of fungal meningitis. Aim: This study was done to improve our current laboratory techniques for the diagnosis of the most important infectious agent causing acute meningitis in our patient population, C. neoformans, because it is undiagnosed in Sudan. Materials and Methods: A cross sectional study conducted in Khartoum state, during period from November 2019 to February 2021. 50 CSF samples were collected from patients with acute meningitis. DNA was extracted, and subjected to conventional PCR test to detect C. neoformans in each sample, data were analyzed using SPSS. Results: The study showed that (10% ) 05/50 of the study populations were detected with C. neoformans, their CNS contains genome of C. neoformans, when using conventional PCR, (6%) 3/05 were males, and (4% ) 02/05 were females, were within age( 28%) 14/50 range 0-30 days (neonate), (56%) 28/50 were within age range 1-12 months (children), while the rest (16%) 08/50 were within age range 13-24 months (children). with insignificant statistical difference p.value = 0.92, 0.84 respectively. Conclusion: In conclusion, this study reveals high prevalence of C. neoformans among patients attending Omdurman Children's Hospital in Khartoum State.
